Sirtuins (SIRT1–SIRT7) are a family of regulatory proteins that “tune” gene activity, metabolism and a cell's stress resistance. Their distinctive feature is dependence on the coenzyme NAD⁺: without it sirtuins do not work.
Sirtuins have become stars of the “science of longevity”: they are linked to the effects of calorie restriction and to the activation of autophagy. But there are also many marketing myths around them — worth separating from the real data.
How sirtuins relate to aging
Sirtuins are activated during an energy deficit (when NAD⁺ levels rise) and help the cell survive stress: they repair DNA, improve the function of the mitochondria and reduce inflammation. Much of the benefit of calorie restriction in model organisms is realized through them.
With age NAD⁺ levels fall, and sirtuin activity declines — one of the reasons for the interest in ways to maintain NAD⁺.
Resveratrol, NMN and the honest picture
Here caution is especially important. Resveratrol (from red wine) was marketed as a “sirtuin activator,” but its direct effect on SIRT1 is disputed, and it has shown no benefit for human longevity. NAD⁺ precursors (NMN, NR) do raise NAD⁺ in the blood, but there is as yet no proof that this extends a human life — only early studies of individual markers.
What works for sure: physical activity and calorie restriction naturally support NAD⁺ and sirtuin activity — without expensive supplements of unproven effect.
