The cellular "energy sensor" that switches on when energy is scarce

AMPK — the cell's energy sensor, an activator of cellular repair

AMPK (AMP-activated protein kinase) is the cell's “energy sensor.” When the cell is low on energy, AMPK switches on and puts it into conservation and repair mode: it activates autophagy, improves the function of the mitochondria and slows growth.

In essence AMPK is the “counterweight” to the satiety sensor mTOR. Its activation is linked to the beneficial effects of fasting and physical exertion and is considered one of the paths to healthy longevity.

What activates AMPK

AMPK switches on wherever energy is spent and the cell's “fuel” drops. The main natural activators are: physical exertion (especially endurance exercise), calorie restriction and fasting, a glucose deficit. Among drugs it is activated by metformin — much of its studied “geroprotective” properties are linked to this.

When activated, AMPK launches cleanup (autophagy), stimulates the formation of new mitochondria and increases sensitivity to insulin — that is, it improves metabolism.

What is important to understand

“Boosting AMPK” with supplements and “activators” is largely marketing: the most reliable activators are movement and moderation in food, not dietary supplements. Metformin is being studied as a longevity agent (the TAME trial), but taking it in healthy people without diabetes is not recommended — there is as yet no proof of longevity benefit in humans.

Practical takeaway: regular aerobic exercise and avoiding constant overeating are the most well-founded way to support this pathway.

Should I take metformin for longevity?

For healthy people without diabetes — no. Metformin activates AMPK and is being studied as a means of slowing aging (the TAME trial), but there is as yet insufficient proof of longevity benefit in humans, and uncontrolled use is unsafe.

Are AMPK and mTOR the same thing?

No, they are two opposing sensors. mTOR responds to an abundance of nutrients and switches on growth, while AMPK responds to a shortage of energy and switches on conservation and repair. A healthy balance between them is an important part of the mechanisms of longevity.
