Autophagy — a cell recycling its damaged components

Autophagy (literally “self-eating”) is the process by which a cell recycles and disposes of its own “junk”: damaged proteins, worn-out mitochondria and other components, reusing their parts.

This is a built-in system of cellular cleanup and repair, and maintaining it is one of the mechanisms of longevity. Autophagy is switched on by an energy deficit: it is activated by fasting and exertion through the sensors AMPK and mTOR.

Why autophagy matters for longevity

With age autophagy weakens, and damaged components accumulate in cells — this reduces their performance and contributes to aging and disease (neurodegeneration, metabolic disorders). In animal studies enhancing autophagy extended lifespan, while blocking it abolished the benefits of fasting and of a number of substances.

Put simply, autophagy is the cell's “maintenance”: the better it runs, the longer the cell stays healthy.

What triggers autophagy

The main physiological trigger is a shortage of energy and nutrients. Autophagy is enhanced by: fasting and calorie restriction, long gaps between meals, physical exertion, and also sleep. It is also activated by the drug rapamycin (through inhibiting mTOR).

This works through the balance of sensors: when energy is short, AMPK switches on and mTOR activity falls — and the cell shifts from “growth” to “cleanup.”

What is proven and what is not

Be careful with promises. Convincing data on life extension through autophagy come mainly from animals; there is as yet no direct proof that a particular fasting scheme extends a human life. Fashionable “autophagy protocols” and “inducer” supplements are overrated.

What is sensible and safe: regular physical activity, not overeating, reasonable gaps between meals. Strict fasting is not for everyone and requires caution (pregnancy, diabetes, eating disorders are contraindications).

How many hours should I fast for autophagy?

There is no exact “activation threshold” for autophagy in humans tied to a specific number of hours — the popular figures (16, 24 hours) are largely marketing. Autophagy increases gradually with an energy deficit, and it is triggered not only by fasting but also by physical exertion.

Does fasting extend human life?

There is as yet no direct proof that a particular intermittent-fasting scheme extends human life — most data come from animals. In humans, calorie restriction improves a number of metabolic markers, but that is not the same as proven longevity.

How can I enhance autophagy safely?

The most well-founded and safe ways are regular physical activity, avoiding overeating and reasonable gaps between meals. Strict prolonged fasting is not for everyone and is contraindicated in pregnancy, diabetes and eating disorders.
