How stress accelerates aging

Chronic stress sets off three interlocking mechanisms, and each one speeds up epigenetic aging. The MIDUS study showed they can be measured through DNA methylation (GrimAge, DunedinPACE), with sleep, activity, and diet acting as the key mediators. The epigenetics of stress: what the study measures

Researchers at Georgetown University used three generations of epigenetic clocks:

GrimAge
The most accurate predictor of mortality. R²=0.43 — 43% of the variation is explained by stress and lifestyle. Stressors acted both through behavior and directly (cortisol, blood pressure).
DunedinPACE
Measures the pace of aging: a value >1.0 means aging faster than normal. Moderately explained by stress (R²=0.31) and sensitive to short-term change.
PhenoAge
Phenotypic age from blood biochemistry. Less explained by stress (R²=0.07) — blood chemistry is less sensitive to stress than DNA methylation.
Key takeaway
Stressful life events affected GrimAge 2–3× more strongly than perceived stress. Sleep is the strongest behavioral mediator.
